01.Maropitan citrate, also known as CJ-11,972 and marketed as Cerenia(antitonine), was originally developed by Pfizer. The molecular weight of Maropitant citrate is 678.82, and the chemical name is (2S, 3S) -2-diphenylmethyl-N -(5-tert-butyl-2-methoxybenzyl) -3-amino-quinine citrate monohydrate.
02.The synthesis method of maropitan was reported in the literature. The synthesis route of this method requires 8 steps to prepare the free base of Maropitant, which involves the Michael addition reaction involving the difficult to control phenylmagnesium bromide Chemicalbook reagent and Cuprous dimethyl sulfide bromide (CuBr•(CH)S), and the multi-step pressure hydrogenation reaction.
03.The above method of synthesizing maropitane free base not only has a long route and low yield (the total yield of the 8-step reaction is only about 10%), but also involves the use of a variety of precious metals, such as Ti, Pt, Pd, etc., which is not only costly, but also requires a lot of solvents and adsorbents to remove these precious metal residues. Therefore, the development of a new synthetic method of maropitane is particularly important for the industrial production of the drug.
